Kinetic Reactivity

The reactivity of the Cd-beta species was analyzed by studying the reaction with DTNB. The reaction takes place according to the following scheme:

Figure 5. Typical plot for the reaction between 5mM Cd3betaC(red)or Cd3betaN(green)and 0.5 mM DTNB in 5 mM Tris / 0.1 M KCl at pH 7.5 and followed at 412 nm, absorbance corresponding to the TNB formed during the reaction.

The kinetic studies carried out with the synthetic isolated domain of lobster metallothionein indicated that a behaviour comparable with the same domain within the holoprotein structure, as it's represented in its reaction with DTNB. The reaction observed are biphasic for each of the species studied. The overall rate expression for the reaction of the Cd3beta derivatives as in the case of the holoprotein (Cd6MT) is found to be:

rate = {k1f + k2f [DTNB] + k1s + k2s [DTNB]} [Cd3b]
