If you have not used the 600 at all before, login to dexter, either at the console or from any NMRFAM workstation. 2. type 'cd /u/data', and then 'ls' 3. if a directory with your username exists, type 'cddata'. 'pwd' should show that you are in the directory /u/data/[username]/nmr. go to #4. 3a. if no such directory exists, create one: type 'mkdir [username]', and then 'mkdir [username]/nmr'. then type 'cddata'. 4. Make sure you are in the directory '/u/data/[username]/nmr' (type 'pwd'). Copy the setup600.tar archive to this directory (this is your working directory on the DMX-600) with the command: 'cp ~volkman/dmx600/setup600.tar .' 5. unpack the archive: 'tar -xvf setup600.tar' 6. If you are on the console of dexter (the DMX-600 workstation), type 'xwinnmr' to start the Bruker software, if it is not already running. In the xwinnmr window, type 'dir' to see the list of basic experiments which have been installed in your directory. _________________________________________________ Experiments included in the setup600.tar archive: -- 1D calibration and tuning 1puls - simple 1 pulse expt; use for tuning 1H channel and calibrating 1H pulses. tune15n - tune 15N channel with wobb tune13c - tune 13C channel with wobb tune2h - tune 2H channel with wobb tune31p - tune 31P channel with wobb calib15n - calibrate 15N pulse widths on standard (NAG) sample (peak at ~ 8 ppm) with decp90-type sequence. water suppression via off-resonance 1-1 sequence. calib13cf3 - calibrate 15N pulse widths on standard (NAG) sample (peak at ~ 2 ppm) with decp90-type sequence. water suppression via off-resonance 1-1 sequence. -- Basic 1D and 2D 1H, 15N and 13C spectra 1d_watergate - 1D 1H spectrum with 3-9-19 selective inversion/ watergate-type solvent suppression. Very robust, should require input of only the 1H 90 pulse (p1). hsgp19_n15 - 2D 15N-1H HSQC experiment with 3-9-19 water suppression and water flip-back.
